Outline of Research at the Start	申請者はマウス膠芽腫モデルを用いて、代謝改善薬でCAR-T細胞のエネルギー代謝状態を改善することで、膠芽腫の低酸素な微小環境を克服し、CAR-T細胞の治療効果を高めることに成功した。この発見についてヒト生体内においての再現性を確認すべく、患者検体由来の膠芽腫オルガノイドを用いてCAR-T細胞の不応答性の原因を免疫代謝の観点から解析する。さらに代謝改善薬で前治療したCAR-T細胞を用いて膠芽腫オルガノイドの治療を行うことでヒト生体内に近い状態で代謝改善薬の影響を確認することができる。本研究を通じて、代謝改善薬によるCAR-T細胞の前治療を膠芽腫に対する臨床応用に近付けることを期待する。
