Outline of Research at the Start

Matrix metalloproteinase（MMP-2）遺伝子の不活性型ミスセンス変異は、ヒトにおいて多関節の関節破壊と骨粗鬆症を呈する多中心性骨溶解症（Winchester症候群）を生じる。Winchester症候群の病態から、骨・関節組織のコラーゲン代謝におけるMMP-2/MT1-MMPの重要性が示されたが、その病態機序は不明である。Winchester症候群の患者由来iPS細胞を用いて、変異型MMP-2とMT1-MMPの相互作用を検討することで、骨・関節破壊の分子機序を明らかにすることが本研究の目的である。
