Outline of Research at the Start

去勢抵抗性前立腺癌の発生メカニズムは、AR標的治療による腫瘍周囲のアンドロゲン濃度低下に応じたARの増幅やAR遺伝子の変異などの遺伝的構造変化の獲得と、アンドロゲン非依存性増殖に関わる遺伝子変異を当初から持つ細胞の可能性が考えられている。本研究では、アンドロゲン非依存性の増殖に関与する遺伝子を同定するため、LNCaPをアンドロゲン濃度勾配下においた上でCRISPR/CAS9ゲノムワイドスクリーニングを行い、低アンドロゲン条件下で増殖する細胞のゲノム変異を解析、候補遺伝子のノックアウト細胞株を作成し、アンドロゲン非依存性に増殖する分子機構を解明する。
