Outline of Research at the Start

保存的治療に抵抗性を示す難治性メニエール病には、中耳加圧療法や内リンパ嚢手術が行われるが、その作用機序は未解明である。本研究では、①内リンパ嚢開放術中に中耳加圧を行い、内リンパ圧変化から内リンパ管閉塞の有無を評価し、②内リンパ嚢組織の遺伝子発現および免疫組織学的解析により内リンパ吸収不全を立証し、③RNA干渉を用いたモデル動物によりイオン輸送機能不全と水腫形成の関係を検証する。これによりメニエール病の病態形成機構と治療の科学的根拠を明らかにし、各治療の信頼性を高めその適応を最適化していくことが期待できる。
