Outline of Research at the Start

喉頭乳頭腫はヒトパピローマウイルス(HPV)の6型や11型が関与し、一般に手術治療が行われるが再発をくり返す難治性疾患である。抗ウイルス剤であるシドフォビルの注入は、繰り返すうちに耐性を示し、有効性は高いが根治率が低いことが課題として浮上した。本研究では、シドフォビル耐性獲得の分子機構解明、そして耐性克服法の開発を行う。本申請では、まず、治療経過中の腫瘍細胞内HPVコピー数変化と遺伝子変異導入頻度の関連性を検討する。これらの現象に関与する宿主遺伝子として HPV関連中咽頭癌において同様の作用を有するAPOBEC3に着目し、鶏卵法を用いて検証する。
