Outline of Research at the Start

PHA synthase (PhaC) is an enzyme that produces biodegradable plastics (PHAs), with a preference for short-chain substrates over medium-chain ones. However, the structural basis of this specificity is unclear. This project aims to reveal intermediate structures of PhaC with its substrates using X-ray crystallography and cryo-EM. By analyzing homologs from different organisms and performing protein engineering, we aim to uncover key structural features that control substrate preference. These insights will support future enzyme engineering efforts.
