Outline of Research at the Start

KRAS G12D阻害剤に対する初期耐性、ならびに二次変異やバイパス経路等の獲得耐性が懸念され、耐性機序の検索や克服に関する前臨床研究による、初期治療ならびに耐性化後の新たな薬剤治療戦略の開発のために基盤となるデータが必要であるため、本研究ではKRAS G12D変異細胞株に対するmutagenesisによる耐性二次変異スクリーニングや、患者由来のゼノグラフトおよび肺癌オルガノイドを用いた網羅的解析やCRISPRスクリーニング等により、KRAS G12D阻害剤に対するon-target・off-targetの初期・獲得耐性機序とそれらを克服するための薬剤検索を行う。
