Outline of Research at the Start	拡張型心筋症（DCM）は予後不良の心疾患であり、現時点で有効な治療法は心臓移植に限られている。DCMの多くは遺伝子異常に起因することから、ゲノム編集治療が注目されているが、ゲノム編集効率や体内デリバリーに課題がある。そこで本研究では、任意の塩基を精密に編集可能なprime editorに着目し、Cas9ニッカーゼドメインおよび逆転写酵素ドメインを、それぞれ小型タンパクに置換することで小型化を図る。さらに、deep mutational scanningにより編集活性を高精度化し、これを単一のアデノ随伴ウイルスに搭載しDCMモデルマウスにおける治療効果を検証する。
