Outline of Research at the Start

髄芽腫では全脳照射とDNA傷害薬を組み合わせ, 生命予後は改善してきたが, 治療関連の認知機能低下, 二次癌など問題となる. 予後不良例を救済する新規治療戦略, 予後良好例への適切な治療強度の選択が同時に求められている.
申請者らは, Schlafen (SLFN) ファミリーメンバーの 1 つSLFN11が, この層別化に有用ではないかと考え研究を行ってきた. 本研究ではSLFN11低発現となる予後不良例を術後早期に同定, 救済するため, ①臨床検体でのSLFN11評価法の確立, および②マウス同所性xenograft modelを用いたDNA修復阻害療法の開発, を行いたいと考えている.
