Outline of Research at the Start |
This project develops a ribosomal system to incorporate γ-unsaturated amino acids (gAAs) into peptides using flexizyme-charged tRNAs and the FIT system. Synthesized gAA-CMEs with diverse sidechains enable formation of thiol-ether-cyclized macrocyclic peptide libraries. These are screened via mRNA display against targets like Pin1 and HPV16 E6. Outcomes include novel peptide inhibitors, a flexible mRNA display toolkit, and insights into ribosomal tolerance for non-proteinogenic monomers.
