Outline of Final Research Achievements

Genomic imprinting, a transcriptional regulatory mechanism that exists only in mammals, is a phenomenon in which certain genes are mono-allelically expressed only when they are paternally or maternally inherited. We have successfully identified genomic DNA region to which an epigenetic mark is added to distinguish parental origin of certain genes in offspring’s cells, by generating and analyzing gene-modified (transgenic, knock-out, and gene-edited) mouse lines.

Academic Significance and Societal Importance of the Research Achievements

DNA塩基配列以外の情報が親世代から子世代へと受け継がれ、それが子世代の遺伝子発現を変化させる例が知られる。ゲノム刷り込みでは、精子や卵の形成過程でゲノムDNAに付加されるメチル化が、由来する親の性を見分ける唯一の目印であると考えられていた。しかし申請者は、DNAメチル化以外の印も、生殖細胞でゲノムに付加されることを見いだし、本研究において、この未知のエピジェネティック情報が書き込まれる場所を特定し、ゲノム刷り込み分子制御メカニズムの一端を解明した。本成果は、クローンによる動物(優良家畜や希少種)生産の問題解決等につながると考えている。
