Outline of Final Research Achievements |
In the evolutionary biology, it is very important issue to identify genes and mutations that are responsible for the adaptation. We performed a thermal adaptation experiment using the single-stranded RNA bacteriophage Qβ in which the culture temperature was increased from 37.2℃ to an inhibitory temperature of 43.6℃ in a stepwise manner in three independent lines. And we obtained three kinds of mutants that were able to amplify at 43.6℃. In this study, we measured their fitness and infection cycle quantitatively, and identified the responsible genes and substitutions for thermal adaptation of Qβ.
