Outline of Final Research Achievements |
In this study, we tried to develop the exocytosis-like secretion system that secretes intra-vesicular content in response to the recognition of specific molecule. To construct such a system, we tried to introduce Ca channel Orai-1 into a cell-sized giant liposome. Orai-1 was expressed in E.coli and we succeeded in isolation and purification of Orai-1. We tried various ways to reconstitute the channel into the liposome, such as changing phospholipids that form liposomes and changing the preparetion methods of giant liposmes. However, we could not reconstitute enough channels into liposomes.
