Outline of Final Research Achievements |
We aimed to develop a deep proteomics analytical system to identify and quantify complete human proteome expressed in human cells. We developed ultrahigh resolution LC-MS systems using meter-scale monolithic silica capillary columns in combined with meter-scale HILIC columns for fractionating or slicing samples. As a result, when using the slicing mode, the number of identified proteins increased by 46%, compared with the conventional method, Further, using the fractionation method, we successfully identified 10,000 human proteins with 200-fold sensitivity and 6-times throughput.
