Outline of Final Research Achievements |
First, we tried to collect neutrophils from normal human peripheral blood. However, there were some technical difficulties about collecting naive neutrophils from human blood. Therefore, we tried to obtain mouse naive neutrophils from bone marrow and analyzed ROS production, NETs release and antimicrobial peptide gene expressions under some stimulative conditions. We successfully isolated naive neutrophils from mouse bone marrow and visualized NETs release under C.albicans stimulation. We will establish the new method for visualizing NETs release and perform gene-based analysis of NETs-associated DNA in autoantibody-mediated diseases in our future project.
