To elucidate the interaction of cells which controls bone homeostasis, the environment in living body is reconstructed outside of the body, and a novel system was established to analyze it using 2-photon microscopy. By culturing osteoblasts which form bones for long-term, and by observing same regions temporary, phenomena occurring during bone modeling such as increase of matrix, appearance of osteocytes which control bone metabolism, and changes in morphologies of osteoblasts on the matrix were reproduced and proved quantitatively. In addition, in co-culture of long-term-cultured osteoblasts and osteoclasts which resorb bones, phenomena in bone remodeling such as matrix resorption by osteoclasts, appearance of mature osteoblasts in the resorption pits and replenishment of resorption pits could be captured at cellular level and quantified.
