Outline of Annual Research Achievements

様々な環境変化に対するサンゴと褐虫藻の応答を正確に評価するためには、その共生メカニズムについて正しく理解する必要があるが、細胞内共生という代謝プロセスの複雑さゆえに、十分な研究が進んでいないのが現状である。本研究の目的は、造礁サンゴとその細胞内に共生する褐虫藻との間の複雑な代謝構造を、炭素・窒素安定同位体トレーサー法を用いて解析することである。
造礁サンゴ－褐虫藻共生系は主に二つの栄養源を持つ。一つは褐虫藻の光合成で生産される有機物、もう一つはサンゴによる動物プランクトンの捕食である。平成26年度はまず前者によって獲得された炭素・窒素の代謝に注目した実験を行った。沖縄本島周辺海域に多く生息するPorites cylindricaとMontipora digitataを実験対象としてそれぞれ25片程度採取し、琉球大学熱帯生物圏研究センター瀬底研究施設内の流水型水槽で数日～数週間養生させた。実験開始時に13Cで標識された炭酸水素ナトリウムと15Nで標識された硝酸カリウムを閉鎖系の飼育水槽に添加し、その中で対象となる造礁サンゴ片を24時間培養した。その後、サンゴは採取した海域に戻し、0、2、10、20、30日目に回収した。回収したサンゴの有機組織は遠心分離器で褐虫藻と動物体サンゴに分け、それぞれの画分に含まれる有機炭素・窒素の安定同位体比を測定した。測定の結果、褐虫藻の窒素安定同位体比は時間経過とともに徐々に減少するが、動物体サンゴでは徐々に増加し、両者の値は近づく傾向を示した。これは窒素が共生系内で循環していることを示唆する。一方、炭素安定同位体比は両方の画分で減少傾向を示し、窒素とは異なる代謝経路を持つと考えられる。今後はこれらの代謝速度を詳細にモデル化することを試みる。
