Outline of Annual Research Achievements

本年度は、昨年度におこなった有機リン系コリンエステラーゼ阻害薬誘発性けいれん波モデルに対するアトロピン（ムスカリン受容体遮断薬）の治療効果に着目し、その責任受容体を解明するために、選択的ムスカリン型アセチルコリン受容体（mAChR）拮抗薬の治療効果の検討をおこなった。ピレンゼピン（M1-mAChR拮抗薬）は、パラオキソンによるPS amplitudeとfEPSP slopeの低下を防止した。p-fluorohexahydro-sila-difenidol hydrochrolide（p-HHSiD：M3-mAChR拮抗薬）は、パラオキソンによるPS amplitudeの低下を防止したが、fEPSP slopeの低下は防止できなかった。メトクロピラミド（M2-mAChR拮抗薬）およびトロピカミド（M3-mAChR拮抗薬）はパラオキソンによるPS amplitudeとfEPSP slopeの低下を防止できなかった。ピレンゼピンおよびp-HHSiDは多発性スパイクの発生率とスパイク数を減少させたが、メトクロピラミドおよびトロピカミドは減少させなかった。
M1およびM3 mAChRはGq/11タンパク質共役型受容体であり、この受容体の活性化はシナプス伝達を促進すると言われいている。有機リン誘発性てんかんは過剰なアセチルコリンの蓄積によるGq/11タンパク質共役型受容体刺激によって誘発される可能性が示唆された。
