マイクロRNA発現解析に基づいた新しい肺移植後閉塞性細気管支炎治療法の確立

Outline of Annual Research Achievements

肺移植後慢性拒絶反応による閉塞性細気管支炎に関与する転写調節因子を検索し，新規治療展開への基盤を確立する目的で以下の実験を行った．
BALB/Cマウスをドナー，C57BL/6マウスをレシピエントとするMHC完全不適合の組み合わせで，異所性気管移植を行なった．この異所性気管移植モデルにおいては移植後経時的に気管上皮の障害が出現し，線維芽細胞増殖により気管内腔が閉塞されることが明らかになっている．実験群，および対照群は，異所性同種気管移植群：BALB/C → C57BL/6，および異所性同系気管移植群：C57BL/6 → C57BL/6とした．移植後7日目，21日目にマウスを犠牲死させ，移植気管を摘出した．
先ず，移植気管組織からRNAを抽出しマイクロRNA（miRNA），およびメッセンジャーRNA（mRNA）のマイクロアレイ解析を行った．miRNAのマイクロアレイ解析により異所性同種気管移植後21日目において亢進していた3種類の遺伝子，抑制されていた16種類の遺伝子が明らかとなった．これら19種類のmiRNAについてリアルタイムPCRを行い，その発現の程度を定量的に評価したところ，異所性同種気管移植後21日目の群で，miR-204-5pがその他の群と比較して有意に抑制されていることが明らかとなった．
次いで，パスウェイデータベースを使用してmiR-204-5pの標的となるmRNAの探索を行った．この結果とmRNAアレイデータを照合し，発現関係がmiRNAとmRNAで逆となっている遺伝子，つまり亢進している20種類のmRNAが明らかとなった．以上の結果の妥当性はクラスター解析でも確認された．
さらに，気管組織の病理組織学的評価（線維芽細胞増殖による気管内腔の閉塞の程度）を行い，miR-204-5pのリアルタイムPCRデータとの関係を解析したところ有意に相関していることが明らかとなった．
