| Outline of Research at the Start |
本研究では、胆管結紮（BDL）マウスモデルを用い、RICの胆汁鬱滞性肝障害に対する短期・長期的肝保護効果とその機序を明らかにすることを目的とする。本研究は３年間を予定する。初年度は、sham群、BDL群、BDL+RIC群を対象にし、術後72時間での微少循環血流、炎症性サイトカイン、eNOSおよびiNOSの影響、酸化ストレス、線維化関連因子の変化を解析し、急性期作用機序を検討する。次年度は、同プロトコールを延長し、術後2週間での肝線維化評価を含め、慢性期での保護効果を評価する。最終年度は、IL-10ノックアウトマウスを導入し、慢性期におけるRICの効果のIL-10依存性を検証する。
