Research Abstract |
In order to analyse the immunoreguratory system involved in the pathogenesis of immune disorders, human lymphocytes from normals, immunodeficiency patients and autoimmune diseases were studied in vitro. 1. An experimental system to analyse antibody production from the lymphocytes stimulated by specific antigens such as ovalbumin,tetanus toxoid,thyroglobulin and acetylcholine-receptor was established. 2. At least two distinct T cell subsets were proved to be involved for the regulation of antibody formation and both antigen specific and nonspecific suppressor factors were secreted from T cells. 3. T cells from healthy individuals suppressed the autoantibody formation from the lymphocytes of chronic thyroiditis or myasthenia gravis. 4. In stimulation with excess dose of thyroglobulin. T cells from healthy individuals secreted humoral factors suppressing anti-thyroglobulin antibody formation from the lymphocytes of chronic thyroiditis. 5. Such T cells were fused with an established T cell leukemia cell line. Thus obtained hybridomas were cloned and one clone which secretes antigen specific suppressor humoral factor for anti-thyroglobulin antibody formation was obtained. 6. Humoral factors which induce suppressor T cells were demonstrated in the sera from cancer patients. Alpha-fetoprotein also induced suppressor T cells.
