Research Abstract |
1. Study for enzyme immunoassay of microorganisms: Antiserum to Fuzarium, a group of fungi which causes various plant diseases, was prepared and used for developing novel enzyme immunoassay of Fuzarium, which enable to apply 12 different spieces of the fungi. Sensitivity of the assay was 1 ng/tube and was applied to measure growth of Fuzarium in media. 2. Enzyme immunoassay of chromomycin A-3 and olibomycin. Highly sensitive enzyme immunoassay for chromomycin A-3 and loibomycin were developed by using the both immune reagents, the antiserum specific to chromomycin and B-D-galactosidase-labeled chromomycin prepared by us. The assay was applied to measure rat serum level of olibomycin, injected with the dose of 2 mg/kg. 3. New method to measure antigen-antibody binding constant. A new method for measuring the binding constant, by the use of enzyme immunoassay with easy procedures, was developed. Theoretical consideration of the method gave a new terms of affinity average binding constant. The binding constant of anti-blasticidin S against blasticidins was measured and the antibody appeared homogeneous judging from the binding constant. 4. Two monoclonal antibody to viomycin were prepared. Both monoclonal antibodies were found to possess affinity only to viomycin molecule, judging from the affinity binding constant values.
