| Research Abstract |
In order to examine if hairy cell leukemia (HCL) in Japanese patients has distinct clinical, hematologic, and immunologic features to qualify a separate treatment as a Japanese variant of HCL, we made following studies; <I> . careful observation of individual patients, <II> and <III> . studies of immunophenotypes and anti-retroviral antibodies in series of HCL patients, <IV> . in vitro experiments using hairy cells, and <V> . application of anti-hairy cell antibody 4B122 to studies <II> and <III> described above (for 4B122, refer to Katayama I. Production of Anti-Hairy Cell Antibody and its Application to Classification of Malignant Lymphomas. Report on the Research Project by Grant-in-Aid for Scientific Research. March 1984).
<I> . During the 3 years of this project, we saw 11 patients with HCL and several patients with closely related hematological disorders. Two of these patients were reported in "A case of prolymphocytic leukemia with ribosome-lamella complex. Nikketsu Kaishi 47:567

, 1984", and "Remission of a CLL variant by splenic irradiation. Nikketsu Kaishi 49:249, 1986 ". These cases showed features intermediate between HCL and PLL (Catovsky D. Semin Oncol 11:362, 1984). Prevalence of these intermediate cases appears highly characteristic of Japanese variant HCL.
<II> . The majority of Japanese HCL patients were phenotyped as HLA- <DR^+> <CALLA^+> <B1^+> <B2^-> <FMC7^+> <Leu1^+> <Tac^+> <alpha> <HC1^-> <alpha> <HC2^-> <4B122^+> . We conclude that <CALLA^+> <Leu1^+> helps to distinguish Japanese from non-Japanese HCL (Leukemia, in press).
<III> . Western blot analyses revealed cross-reacting antibodies to both HTLV- <I> and HTLV- <II> in sera from 10 of 11 patients with Japanese HCL, suggesting an interrelationship between retrovirus and Japanese HCL (Leukemia, in press).
<IV> . In vitro experiments using hairy cells have not yet produced cell surface changes which distinguishes hairy cells from other leukemic or normal lymphocytes.
<V> . 4B122 was tested along with sera from HCL patients in studies <II> and <III> listed above, which found 4B122 to be positive in all HCL and negative in all but one lymphoma cases, and 4B122 to be reactive with p21 of HTLV- <II> by Western blot analysis (both results are in Leukemia, in press).
