Research Abstract |
The experimental studies using rats were designed to investigate; (1)the effect of vascularity on autologous and allogeneic joint transplantations, (2)the influence of major histocompatibility antigen on survival and functioning of grafted joints. Knee joints from 5 strains of the inbred rat were used in our study. The rats were classified into 2 groups. I:Vascularized autologous joint transplantation (from WKA to WKA rats) II:Vascularized allogeneic joint transplantation. Group II was further classified into 4 subgroups according to the difference of haplotypes (subregions) of rat major histocompatibility (RT1) antigens between the donor and recipient rat:1)RT1-A.B.D. barrier (from WKA to LEJ rats), 2)RT1-B.D. barrier (from TO to LEJ rats), 3)RT1-A. barrier (from BUF to LEJ rats), and 4)non-RT1 barrier (from WKA to W rats). Roentogenograms and 99m-Tc scintigrams were taken every 1 week after joint transplantation to investigate the time of bone healing and revascularization of transplanted joints. Histological studies were also performed. Roentgenograms revealed that the bone healing began at 2.6 weeks post-operatively in Group I and Group II-4). The periosteal reaction of the transplanted joints wan not observed until 10 weeks post-operatively in Group II except in subgroup 4). Scintigrams showed the immediate vascularity of transplanted joint in Group I and Group II-4). The vascularization of transplanted joint was observed until 2 weeks post-operatively in Group II-1), 2), and 3to 4 weeks in Group II-3). However, in Group II except in subgroup 4) bone, articular cartilage and surrounding muscle tissue became to be necrosis. These studies suggest that the success of the vascularized allogeneic joint transplantation is dependent on the vascularity and low antigenicity of transplanted joint.
