Research Abstract

The protein contained in gingival fluid was clearly fractionated into 35 bands by agarose-gel isoelectric focusing using Ampholine ( pH 3.5 - 9.5 ) and silver staining. Ten proteins out of these 35 were identified immunologically. In the case of gingivitis and periodontitis, the number of protein bands increased, particularly at around pH 6.6 and pH 8 - 9.as the symptoms aggravated. The proteins increased in amount at the alkaline pH range were immunoglobulins. Further attempts of fractionation using a high performance liquid chromatograph ( HPLC ) presented 12 protein fractions. Since this HPLC method was highly reproducible, we believe that this will serve as a very useful technique for isolating proteins and for subsequent analysis of individual proteins.
Measurement of pH using a transistor pH elctrode showed that the pH of the periodontal pocket of patients with periodontitis was more acidic campared with that of the gingival sulcus of normal subjects, in particular the glossal sid … More e exhibited the mean pH of 6.44 <+!-> 0.48 (X <+!-> S.D.) which was significantly ( p<0.05) smaller than the value of 6.79 <+!-> 0.39 for normal subjects. It was also found that the pH of the the gingival crevices became more alkaline as the volume of effusion increased, and that the buccal side and proximal surface showed constant pH values regardless of the volume of fluid, while the pH of the lingual side decreased with the increase of the fluid volume. The activities of LDH, its isozymes and <beta> -glucuronidase, proteins contaioned in gingival fluid, were assayed by an assembled assay system. It was found that the activity of LDH tended to increase as the degree of infiammation advanced, while the fraction of <LDH_5> was consistently the main fraction of LDH isozymes indicating no change in the pattern. Following to LDH, <beta> -glucuronidase activity also reflected theextent of inflammation.
Clinically, the mobility of abutment teeth and the alveoler bone resorption were examined in patients with the distal-extention removable partial denture, to whom a rigid support type, namely cone telescopic denture was applied ispilaterally and a stress breaking type, namely mini dalbo attacnment denture was attached contralaterally, and it was found that the stress breaking type denture showed not only greater mobility but also a tendency of larger resorption of the alveolar bone.
