Research Abstract

This study was done in order to establish more objective criteria of the treatment effects of radiotherapy or chemotherapy. For this aim, we applied a semiautoscanning microphotometry system connected to a computer with home made software. We characterized the nuclear atypia of cells for oral cancers during the course of the treatments. We measured the nuclear DNA by this system in order to judge the effect of radiotherapy on DNA. Then we measured an effective nuclear area, the total quantity of DNA and the average density of this quantity of DNA. In addition, we caliculated the average DS (Discriminant Score) and MGD (Malignant Generalized Distance). It is important to investigate how these parapeters changed with increasing doses of irradiation. These parameters peaked at 10Gy irradiation, and later showed a tendency to be similar to the quantity of normal cells. During the course of radiotherapy, we noted that the average DS was positive, and that it came comparatively close to a normal level. We also investigated how small the MGD was. As the fundamental examination of chemotherapy, we investigated various parameters of cell kinetics by using human salivary gland tumors in nude mice with this system and with [ <^3H> ]-thymidine autoradiography method. We investigated disturbed tissue or cells that were effected by antineoplastic (VCR) and their recovery process with these parameters. Our results showd that even if the tumor had a sesitivity to chemotherapy, it had a repropagative tendency after two or three weeks, and that the parameters in cell kinetics had the same tendency.
