Research Abstract |
An automated direct assay system using high performance liquid chromatography with electrochemical detection and ultraviolet spectrophotometric detection was developed for the simultanltaneous measurement of estradiol, estrone, progesterone, 17alpha-hydroxyprogesterone, 20alpha-hydroxyprogesterone, testosterone and androstenedione in biological fluids. It takes 30 min to measure estrogens, and 80 min to measure whole steroid hormones. The recovery rates of steroid hormones were 77-84%, and the intra-assay coefficients of variation of the peak height were 1.7-3.1%. The relationship between the dose of each steroid hormone and the peak height showed good linearities. A comparison between the values measured by this method and by radioimmunoassay revealed good correlation for estradiol (r=0.938, p<0.001) and progesterone (r=0.903, p<0.001). Estradiol and estrone could be analysed above the level of 250 pg/ml, andprogesterone, 17alpha-hydroxyprogesterone, 20alpha-hydroxyprogesterone, testosterone and androstenedione could be analyzed above the level of 5.0-7.5 ng/ml. The method was applied to the clinical appraisal of placental function and maturation of ovarian follicles.
