Research Abstract

Studies on the genome extraction of the DD genome plant from a wild rice, Oryza alta (2n=48,genome CCDD) and of theBB genome plant from emmer wheat, Triticum durum (2n=28, genome AABB) were carried out using the techniques of tissue culture and p-fluorophenylalanine(PFP) treatment.
In the case of a wild rice, triploid plants (2n=36, genome CCD or CDD) were obtained among plants regenerated from callus culture of young florets of O. alta (2n=48, tetrapolid plant) on the medium containing 50 mg/l of PFP. Trials to obtain the CC or DD genome plant from these triploid plants have not been succeeded so Far.
In the case of a emmer wheat, a haploid plant (2n=14, genome AB) were picked out among plants from seeds obtained by injection of PFP solution into glumes of T. durum (2n=28, tetraploid plant) at 6 hours after fertilization. Callus cells from this haploid plant after PFP treatment contain reduced number of chromosomes less than haploid number, but A or B genome plant have not been obtained so Far.
