Research Abstract |
Epithelia cells from a neonatal rat tougue were sucessfully grown in cell culture The isolated epithelial cell culture is decsribed in association with the patterns of growth and maturation differences, formation of keratohyalin granules and keratinization. The cell doubling required about 30 h.. Maturation and vertical stratification of the culture cells occurred at a later stage, demonstrated by an increase in cell organelle, tonofilament accumulation and present of keratohyalin granules. No membrane coating granules were detectable. In replicas, density and size of gap junction and desmosome increased with proliferation, differentiation and maturation of the cells. Treatment with DMBA caused hysiological and morphological changes. Transient decrease of membrane resistance and potential occurred prior to the decrease in coupling ratio at 6 h. after treatment with DMBA. In replicas, at 6 h. after treatment with DMBA, the number of intramembranous aprticles(IMP) was less than that of the control. At 12 h, the number ofIMP increased by 20-40% more than the control. At 3 day, the number of IMP decreased again and remained about the same as that of the control. The number of gap junctions decreased by 45% of the control value at 6 h. after treatment with DMBA. At 12 h and thereafter, the number and area of gap junctions subsequently decreased by 60-80% of the control value. Alteration in number and area of desmosomes were similar to those of gap junctions.
