Research Abstract |
The prism sheath of porcine immature enamel consists of high molecular weight proteins(higher than 30000 daltons) and highly aggregative lower molecular weight components(in the 6000-30000 range). From these components, glycine and tyrosine rich components(MW approximately 7000) and serine and glutamic acid rich components(MW approximately 12000) were separated by gel filtration, and were purified by HPLC using C-18 reverse phase column. The results of amino acied analysis and amino acid sequencing indicated that glycine and tyrosing rich component was derived from amelogenin and it corresponded to approximately one third of the molecular length from the amino terminal of the molecule. On the other hand, as indicated below, the sequences of the first 20 residues of two serine and glutamic acid rich components were different from that of amelogenin. 1. Met-Leu-Ser-Gln-Tyr-Ser-Arg-Phe-Gly-Phe-Gly-Lys-Ser-Phe-Asn- Ser-Leu-Met-Ser-Ser- 2. Met-Lue-Ser-Gln-Tyr-Ser-Arg-Phe-Gly-Phe-Gly-Lys-Ser-Phe-Asn- Asp-Leu-Asp-Met-Ser-
