Research Abstract |
In order to obtain a clue to know the control mechanism of phasic and tonic <alpha> -motoneurons innervating fast and slow muscles, a series of studies was carried out and following results were obtained. 1. <alpha> -Motoneurons received inputs from substance P (SP), serotonin (5-HT) and enkephalin (Enk)-containing presynaptic boutons. The SP-immunoreactive presynaptic bouton contained spherical dense-cored vesicles and clear vesicles, 5-HT-immunoreactive bouton elongated dense cored vesicles and flattened clear vesicles, and Enk-immunoreactive bouton spherical dense cored vesicles and clear vesicles of mixed types. 2. Acetylcholinesterase activity was observed in the synaptic cleft of the two types of presynaptic boutons: (1) S type bouton; (2) boutons containing spherical dense cored vesicles and spherical clear vesicles. 3. <alpha> -Spectrin was purified from chicken red blood cell ghosts and specific antibody against <alpha> -spectrin was raised in rabbits. , the localization of <alpha> -spectrin was studied immunocytochemically in the chicken and monkey ventral horns. <alpha> -Spectrin-like immunoreactivity was observed in cortical cytoplasm of the soma, dendrites and axon of large ventral horn cells. The postsynaptic density was particularly intensely immunostained. The immunostaining was also associated with neurotubules in the axon and dendrites. 4. The SP-like immunoreactive structures were studied in the laminae I and II of the chicken dorsal horn. Two kinds of immunoreactive boutons were observed: (1) the immunoreactive presynaptic boutons showing approximately circular profile which synapsed with unstained dendrites; (2) the synaptic glomerulus in which a few immunoreactive axon terminals were surrounded by several axons and dendrites. In the latter, central axon terminals constituted presynaptic components in most synapses but in a few postsynaptic components. Septate junction-like structures were observed between individual central axon terminals. 5. Quantitative analyses of the synaptology of phasic and tonic <alpha> -motoneurons were performed. The ratios of the number of Gf-type boutons (PLD>ALD) and S type boutons (PLD<ALD) to the total number of boutons were significant.
