Research Abstract |
1. Establishment of galactose oxidase-Schiff (GOS) reaction : All of commercially available galactose oxidase contained contaminations which showed protease activity. Two years experiments demonstrated that an affinity colum chromatography with G-200 was most effecient to remove contaminated materials. For the histochemical use, however, removal of protease activity was achieved by incubating an enzyme solution at 50C for 30 min. Oxidation with this enzyme at the concentration of 50-100 units/ml for 8 hr at 37C was sufficient to obtain consistent results. Engendered aldehyde groups could be visualized by ordinary Schiff reaction, but more effectively visualized by black Schiff reation or cold thionin Schiff reaction. 2. Application of GOS reaction : In the gastrointestinal tract, the surface mucous cell of the stomach showed most intense reaction. Gastric carcinoma of either differentiated or undifferentiated type unexpectedly contained surface mucous cell type-carcinoma cells. Studies of early gastric type of differentiated type revealed that carcinoma tissues proliferated in the mucosa showed laminated structure which consisted of covering epithelial cell-type carcinoma cells in the upper layer and glandular epithelial cell-type carcinoma cells in the lower. This organoid differentiation possively mininizing the invasion beyond the muscularis mucosa. GOS reaction was also applied to analyse stored materials of GM1 gangliosidosis and demonstrated as a useful technique to determine sugar structures in macrophages and various epithelial cells.
