Research Abstract |
THE ACTIVITY OF F1 ANTI-PARENTAL AUTOREACTIVE CYTOTOXIC T LYMPHOCYTES (CTL), GENERATED BY CULTURING THE SPLEEN CELLS (SPC) OF 70% HEPATECTOMIZED (BALB/c X C57BL/6) F1 (CB6F1) MICE WITH PARENTAL C57BL/6 SPC, DECREASED TEMPORARILY AND REACHED THE LOWEST LEVEL FOUR DAYS AFTER THE HEPATECTOMY. THE REDUCTION WAS CAUSED MOSTLY BY SPLENIC ADHERENT CELLS (SAC), AND WAS ABOLISHED BY INDOMETHACIN. WHEN THESE HEPATECTOMIZED F1 MICE WERE INJECTED WITH UNTREATED PARENTAL C57BL/6 SPC FOUR DAYS AFTER THE OPERATION, TOLERANCE NOT ONLY TO THE INJECTED PARENTAL CELLS BUT ALSO TO THE OTHER PARENTAL CELLS CONTINUED FOR AT LEAST FOUR WEEKS AFTER THE INJECTION. THIS TOLERANCE TO THE PARENTAL CELLS WAS NOT LOST AFTER THE ADDITION OF INDOMETHACIN AT THE INITIATION OF THE MLR CULTURE. THESE TOLERATED F1 MICE, HOWEVER, PRODUCED CTL AGAINST THIRD PARTY CELLS TO THE SAME EXTENT AS THE CONTROL MICE. IN ORDER TO STUDY THE MECHANISM OF AUTOREACTIVE CTL TOLERANCE, LIVER CELLS WERE CULTURED WITH SYNGENEIC SPLENIC T CELLS. MMC TREATED NONPARENCHYMAL PLASTIC ADHERENT LIVER CELLS (CONSISTING MAINLY OF KUPFFER CELLS) STIMULATED THE PROLIFERATION OF SPLENIC T CELLS. HOWEVER, THE KUPFFER CELLS SYNGENEIC TO RESPONDER CELLS IN MLR SUPPRESSED MLR. THE CULTURE SUPERNATANT OF KUPFFER CELLS ALSO STIMULATED SYNGENEIC SPLENIC T CELLS AND SUPPRESSED MLR AS KUPFFER CELLS DID. ON THE OTHER HAND, MMC TREATED PARENCHYMAL LIVER CELLS AND THEIR CULTURE SUPERNATANT STIMULATED NOT ONLY SYNGENEIC SPLENIC T CELLS BUT ALSO MLR. NOW WE WERE STUDYING THE ACTIVE SUBSTANCES OF CULTURE SUPERNATANT AND THE FUNCTION OF SPLENIC T CELLS ACTIVATED BY THE SUPERNATANT.
