Research Abstract |
Recent studies have shown that antinuclear antibodies (ANAs) in sera of patients with various connective tissue diseases are directed against variety of non-histone nucleoproteins. In double immunodiffusion tests using rabbit thymus extract as the antigen, 472 sera from patients with various collagen diseases were screened for precipitating antibodies to non-histone nucleoproteins. Twenty-three sera(5%) showed undifferentiable precipitine lines. From these precipitating antibodies, a new antigen-antibody system named Fu system was described. The Fu antigen was a non-histone nucleoprotein not containing RNA or DNA. All three patients with anti-Fu antibody were diagnosed as SLE. In immunoprecipitation assay using <^(32)P> -labelled nucleoproteins 341 sera from patients with collagen diseases were screened for antinuclear antibodies against small molecular RNA containing nucleoproteins(snRNPs). The antibodies were detected in 159/341 sera tested. These antibodies could be classified into three types by combination of snRNPs precipitated : anti-U1RNP antibody only, anti-(U1,2,4-6) RNP(Sm) antibody and anti-(U1,U2)RNP antibody. There were 13 sera containing anti-(U,U2)RNP antibody. Patients with the antibody were diagnosed as overlap syndrome in 7, PSS in 5, PM/DM in 1.
