Research Abstract |
1. Colloid-gold labelling of mitochondrial enzymes for immuno-electron microscopy: Ultrastructural localization of mitochondrial electron-transport enzymes, complex <I> , <III> , and <IV> was accomplished by immunocytochemistry on methacrylate embedded muscle sections. Results showed a very distinct and dense labelling of complex <I> , <III> , and <IV> , respectively in the inner membrane of mitochondria. Whereas a few gold particles were present in matrix. Labelling on the paracrystalline inclusions was substantially weaker in abnormal giant mitochondria, but gold particles were extensively confined on the membrane of cristae surrounding the inclusions. 2. Immunocytochemical demonstration of complex <III> and <IV> in tissues from patients with mitochondrial encephalomyopathy: Mitochondria were specifically stained in sections of formalin-fixed paraffin-embedded human tissues by ABC method. A autopsy case with mitochondrial encephalomyopathy showed a deficiency of complex <IV> (cytochrome c oxidase) in skeletal muscle only. 3. Immunocytochemical studies on SSPE virus: Immuno-electron microscopic studies on SSPE virus infected cells and brain tissues were carried out using monoclonal antibodies against measles virus polypeptides. The nucleocapsids in the dendrites of hamster infected with measles virus were stained with monoclonal antibody against NP.
