Research Abstract |
This experiment showed that the characteristic difference in the regulation system of blood flow between normal and malignant tissue was produced by angiotensin II induced hypertension (150 mmHg). Blood flow in tumor tissue of VX-2 carcinoma transplanted intramuscularly in rabbit was remarkably increased by angiotensin II induced hypertension, while blood flow in normal tisse of rabbit (liver, kidney, muscle), was almost constant after induction of hypertension. Based on this findings, a new approach to cancer chemotherapy, combined chemotherapeutic agent and angiotensin II was discussed by using of murine neuroblastoma(C-1300) transplanted intracutaneously in A/J mice. Marked enhancement of the chemotherapeutic effect was noted in the group received active type cyclophosphamide(40487) intravenously under angiotensin II induced hypertension, compared with the control group and the group receiving active type cyclophosphamide(40487) alone intravenously. In next experience, the concentration of active type of cyclophosphamide in blood and tumor tissue were measured sequentially after injection of inactive type of cyclophosphamide intravenously to A/J mice bearing C-1300 neuroblastoma. The peak time which appeared active type of cyclophosphamide in blood and tumor tissue of A/J mice was 60 minutes after the injection of inactive type of cyclophosphamide. This combination chemotherapy of angiotensin II and cyclophosphamide was applied to a 9 years old girl with rhabdomyosarcoma, who had lung metastasis and multiple dissemination of abdominal cavity. Followed this combination chemotherapy of angiotensin II, cyclophosphamide, vincristine and actinomycin D, her abdominal circumference was decreased sooner than after simple type of VAC chemotherapy.
