Research Abstract |
SINCE THE ESTABLISHMENT OF THE TWO STAGE CARCINOGENESIS, VARIOUS TUMOR PROMOTERS HAVE BEEN REPORTED. EFFECTS OF TUMOR PROMOTING AGENTS ( PHORBOL ESTER ) ON THE MALIGNANT TRANSFORMATION OF THE HYDATIDIFORM MOLE CELLS ( BM-26 AND BM-34 CELLS ) MAINTAINED FOR OVER TWO YEARS IN CULTURE, WERE EXAMINED. IN VITRO COLONY ASSAY SYSTEM IN SOFT AGAR AND XENOGENEIC TRANSPLANTATION METHOD WERE USED TO PROVE WHETHER MALIGNANT TRANSFORMATION OF THE HYDATIDIFORM MOLE CELLS TREATED WITH PROMOTER ( TPA: 12-O-TETRADECANOYL-PHORBOL-13-ACETATE ) HAD OCCURRED. THE HYDATIDIFORM MOLE CELLS WERE TREATED WITH TPA ( 0.1, 1.0, 10, 100 NG/ML ) FOR VARIOUS LENGTH OF TIME ( 2, 4, 6, 8 WEEKS ). WHEN THE TREATED CELLS ( 0.2-1 x <10^4> CELLS ) WERE SEEDED IN SOFT AGAR UNDER PRESENT TREATMENT CONDITION WITH TPA , THE PLATING EFFICIENCY WITH OR WITHOUT THE TREATMENT WAS 0.22-2.125 % . SUBSEQUENTLY, THE PLATING EFFICIENCY BY IN VITRO COLONY ASSAY SYSTEM WAS AT FIRST SIGNIFICANTLY INCREASED IN THE CELLS TREATED WITH EACH CONCENTRATION OF TPA FOR EIGHT WEEKS. IN ADDITION, WE DID NOT OBTAIN ANY PROGRESSIVELY GROWING TUMOR EVEN IF WE HAD ATTEMPTED TO TRANSPLANT THESE CELLS INTO NUDE MICE.
