Research Abstract |
BASIC EXPERIMENTS FOR THE FUSION AND DESTRUCTION OF LIPOSOMES WERE CARRIED OUT AS FOLLOWS. 1. A LARGE SCALE PREPARATION METHOD OF PROTEOLIPOSOMES WAS DEVELOPED BY INTRODUCING THE DIALYSIS PROCEDURE. 2. A DYE LASER FLASH PHOTOLYSIS APPARATUS WAS CONSTRUCTEDINCORPORATING INTERFACES FOR A PERSONAL COMPUTOR. A DEVICE CONTROL AND DATA ANALYSES COMPUTOR PROGRAMS WERE PREPARED BY USING C LANGUAGE. 3. SPECTROPHOTOMETRIC AND ELECTROCHEMICAL PROCEDURES WERE DEVISED TO MONITOR THE DESTRUCTION OF LIPOSOMES. 4. WHEN BOTH BACTERIORHODOPSIN AND CYTOCHROME OXIDASE LIPOSOMES WERE FUSED WITH CALCIUM ION, A RELEASE OF RESPIRATION WAS OBSERVED WHEN IRRADIATED. THIS EXTENT WAS UTILIZED TO MONITOR THE FUSION OF LIPOSOMES. 5. LIPOSOMES RECONSTITUTED WITH PROTOPORPHYRIN WAS DESCTRUCTED RAPIDLY WHEN IRRADIATED, ITS EXTENT BEING DEPENDENT ON THE CONCENTRATION OF OXYGEN IN THE MEDIUM. UNDER NITROGEN THE DESTRUCTION WAS INHIBITED TO A LARGE EXTENT. 6. THE ENZYMIC ACTIVITY OF LIPOSOMES RECONSTITUTED WITH CYTOCHROME OXIDASE AND PROTOPORPHYRIN WAS INHIBITED ALMOST COMPLETELY BY IRRADIATION. 7. PHOTOEXCITATION OF PORPHYRINLIPOSOMES AND ITS DECAY WERE ANALYZED BY THE FLASH-PHOTOLYSIS METHOD REVEALING THE BIPHASIC DECAY OF THE EXCITED STATE. THE DECAY WAS ALSO DEPENDENT ON THE OXYGEN CONCENTRATION.
