Research Abstract

骨髄造血微小環境の発生に関する検索では, 胎生18日令マウスの骨髄から培養した壁付着細胞が骨内膜細胞に由来し, 造血支持能を有し, その造血支持能にマクロファージが関与することが示唆された(橋本). 発生学的に骨内膜細胞は前脂肪細胞であると推測されるが(橋本), 林は放射線照射を受けた直後に炭素粒子を静脈注射されたラットの骨髄で, 3日目に炭素粒子が脂肪細胞と骨内膜細胞に摂取されることを認めた.
骨髄の間質細胞の培養細胞株にHー1, KM(張ケ谷), SH(丹下)があり, 今回樹立されたMSー1株(森)はCFU-Sなどの造血幹細胞の増殖分化を長期的に支持する. NIH-3T3線維芽細胞株はILー3無添加で肥満細胞の増殖を支持し, 貧血マウスの肥満細胞はILー3添加によって正常に維持されるが線維芽細胞との接触に依存する増殖に欠陥がある(北村). SH株はヒト巨核球系細胞の顆粒数を増加させる分化因子を出している(丹下). KMー102株が示すBPAは今回の精製蛋白のアミノ酸分析の結果GM-CSFに似ており, 未分化幹細胞レベルに働くことが示唆された(張ケ谷). 前脂肪細胞(Hー1)に対して作られたモノクローナル抗体に認識される細胞接着分子は造血臓器の血管内皮細胞にも依存する(張ケ谷).
血液疾患との関連において, 白血病細胞の増殖には造血微小環境の細胞との接着が必要であることが共生培養系(KMー102およびCA膜)で証明された(張ケ谷, 関). 加令的変化であるin vivoのCFU-Sの回復能の減少はCFU-S自身よりも環境の側の変化による(井上). トロトラスト患者および再生不良性貧血ではCFU-Fの減少とCFU-E, BFU-Eの減少に強い関連性が示唆された(平嶋).
