Research Abstract |
In this project, we examined the vibrational optical activity observed under the existence of second components with the Raman optical activity method (ROA). The ROA is based on the intensity difference of the Raman band excited right and left circularly polarized lights in the case of chiral molecules. We pursued the informations about the structures and biological characteristics. In the first, using cyclodextrin and protein, and azo dyes as chemical proves, we succeeded in the measurements of vibrational optical activity induced by the interaction with the above-mentioned chiral molecules. That is, by the inclusion phenomena between azo dyes cogored and benzopuplin 4B, which are achiralnmolecules, and the abovementioned chiral molecules. ROA signals were quite different between-cyclodextrin and bovin serum albumin systems. This fact shows that the moieties inducing vibrational optical activities seem to be different. In the next attempt, combining SERS(Surface Enhanced Raman Scattering) with ROA, we studied the ROA of amino acids in the silver colloid solution. There is a theory that in the SERS, not only the enhancement of the intensities of Raman bands due to the effect but also the difference between the intensities excited by right and left circularly polarized lights is to be larger, i.e., the intensities of ROA is to be enhanced. However, this theory was not realized till now. We measured several amino acids in the silver colloid solution. As a result, we succeeded to obtain the ROA signal for L-phenylalanine. ROA intensity, <DELTA> ((I^r_-I^l_)/(I^r_+I^l_)). the result present a new methodology for measurements of dilute solutions, though the SERS effect makes <DELTA> values so larger as expected. Further examinations, thoretically and experimentally, are considered to be necessary. However, it is significant to emphasize that the experimental result obtained here is literally the first one not only in Japan but also in the whole world. Less
