Research Abstract

ORNITHINE DECARBOXYLASE (ODC), A KEY ENZYME IN POLYAMINE BIOSYNTHESIS, HAS AN EXTERMELY SHORT HALF-LIFE, IN THE PRESENT STUDY, I INTENDED TO ELUCIDATE THE PARTICIPATION OF ANTIZYME (A POLYAMINE-INDUCED PROTEIN WHICH BINDS TO THE ENZYME AND INACTIVATES IT) AND ANTIZYME INHIBITOR (A PROTEIN WHICH RELEASES ACTIVE ODC FROM ODC-ANTIZYME COMPLEX BY REPLACEMENT) IN ODC DEGRADATION, 1, ANALYSIS WITHATEMPERATURE-SENSITIVE MUTAN CELL LINE DEFECTIVE IN UBIQUITIN CONJUGATION AND FIBROBLASTS FROM A PATIENT WITH I CELL DISEASE INDICATED THAT UBIQUITIN-DEPENDENT OR LYSOSOMAL PATHWAY IS NOT IMPORTNAT FOR ODC DEGRADATION, 2, SIMLAR CROSS-REACTIVITY WITH SEVEN MONOCLONAL ANTIBODIES TO ANTIZYME AND IDENTICAL BEHAVIOR ON ION EXCHANGE AND GEL CHROMATOGRAPHIES OF ANTIZYME FROM HTC AND ITS ODC-STABILIZED VARIANT HMO CELL LINES SHOWED THAT THE INCREASED HALF-LIFE OF ODC IN HMOA CELL IS NOT DUE TO ALTERATION IN ANTIZYME PROTEIN, 3,IN CONTRAST TO PREVIOUS REPORT, MOUSE KIDNEY ANTIZYME IS NOT ONLY INDUCIBLE BY EXOGENOUS PUTRESCINE BUT ALSO PRESENT AS ODCANTIZYME COMPLEX UNDER PHYSILOGICAL CONDTIONS. MOREOVER, CORRELATION BETWEEN ODC DECAY RATE AND ANTIZYME/ODC RATIO IN MOUSE KINDEY SUPPORTS UBIQUITOUS ROLE OF ANTIZYME IN ODC DEGRADATION, 4, ANIIZYME INHIBITOR WAS HIGHLY PURIFIED FROM RAT LIVER AND ITS PROPERTIES WERE EXAMINED, A MONOCLONAL ANTIBODY AGAINST THIS PROTEIN WAS OBTAINED, 5, ANTIPARALLEL FLUCTUATIONS OF ANTIZYME INHIBITOR AND ANTIZYME IN RAT LIVER AND HEART SUGGESTED THAT ANTIZYME INHIBITOR IS NOT A DEGRADATION INERMEDIATE OF ODC BUTA DISTINCT PROTEIN INVOLED IN ODC REGULATION. IT IS LIKELY THAT WHILE ANTIZYME ACCELERATES ODC DEGRADATION, ANTIZYME INHIBITOR AMPLIFIED ODC INDUCTION BY TRAPPING ANTIZYME AND THEREBY SUPPRESSING ODC DEGRADATION.
