Research Abstract |
Hormone synthesis and secretion in pituitary adenomas were investigated by molecular biological techniques. RNA dot, northern blot and in situ hybridization were employed in addition to the immunohistochemistry and endocrinological evaluation of the patients. The results were as follows. 1) PRL and PRL mRNA in adenoma tissue were demonstrated in 5 out of 6 cases of human pituitary adenomas with hyperprolactinemia. In only one case, no PRL and PRL mRNA were detected in the tumor tissue. Hyperprolactinemia in this casc was suggested to be caused by the disturbance of prolactin inhibitory factor by stalk compression. 2) In 18 cases of acromegaly, PRL in the adenoma tissues was demonstrated in 9 cases by immunohistochemistry, while PRL mRNA was detected in 13 cases by cytoplasmic dot hybridization. 3) In 16 cases of clinically non-functioning adenomas, expression of PRL, GH and ACTH gene were studied. PRL mRNA was demonstrated in three cases, and ACTH mRNA in only one case. 5) In the rat pituitary, estrogen stimulates PRL gene expresslon and bromocriptine suppresses it. However, responsiveness to these agents were different from one cell to the other. Thus, the pathophysiology of pituitary adenomas is clarified by the study of gene expression more than the usual approaches.
