Research Abstract |
We have been investigating the biological active diterpenoids from dry leaves of Rabdosia trichocarpa Hara,and determined the structuresof Trichorabdal A,B,C,(TA,TB,TC)and other 5 Trichorabdals,and reported the antitumor activity of them. Now we have found Trichorabdals to suppress the steroid hormone release from isolated rat adrenal cells stimulated with ACTH(ID_<50> 15-50 um).From the the same <alpha>-methylene cyclopentanone system as in antitumour activity,and we synthesized the new suppresser,15-oxohaurenes hydroxylated at C-7,C-9,or C-12. In vivo activity,TB and17-hydroxykaurene increased serum corticosterone in rat and mous and thisdifferent effect from that of in vitro system may acts through the central nervous system. Though TB(30mg/kg,ip)had no effect on blood brain barrier becuse of in vivo steroid hormon stmulatory effect,we are investigating the effect of synthetic diterpenoid which suppress the in vivo steroid hormon release. /Nodosin/15-Oxokaurrene/Hydroxylated 15-Oxokaurene/Steroidogenic actibity
