Research Abstract

我々は染色体ソ-ティングの技術に改良を加え、最近ポリアミン・ジギトニン処理法でヒト培養リンパ球(GM130B)から得た分裂中期染色体を、ヘキスト33258またはヨウ化プロピジウムで染色し、FACS 440セルソ-タ-を用いて合計18画分に分別できるようになった。さらに各染色体画分を直接ニトロセルロ-スフィルタ-に集め、ラベルしたDNAプロ-ブとのスポットブロットハイブリダイゼ-ションを組織的に行う技術も確立した。このようにソ-ティングした染色体と雑種細胞パネルのDNAを併用して、本年度は以下のヒト遺伝子をマッピングした。Calpain subunit1(CAPN1)/第11染色体、同subunit2(CAPN2)/第1染色体、同subunit関連遺伝子(CAPN3)/第15染色体、同subunit4(CAPN4)/第19染色体、Calpastatin(CAST)/第5染色体、水疱性類天疱瘡抗原蛋白遺伝子/第6染色体にマップした。
