DISTRIBUTION OF PROTEASES IN THE HEPATOPANCREAS OF ERIMACRUS ISENBECKII, GERYON AFFINIS,PARALITHODES CAMTSCHATICUS, PARALITHODES BREVIPES, NEOLITHODES NIPPONENSIS, LITHODES AEQUISPINUS, PARALOMIS MULTISPINA, CHIONOECETES JAPONICUS, AND PANULIRUS JAPONICUS WERE INVESTIGATED. HIGH TRYPTIC AND APPARENT COLLAGENOLYTIC ACTIVITY WERE DETECTED IN ALL SPECIES WHEREAS CHYMOTRYPTIC ACTIVITY WAS UNDETECTABLE. THERE WAS NO EVIDENCE OF ZYMOGEN OF ANY PROTEASE. BY GEL FILTRATION AND ION-EXCHANGE CHROMATOGRAPHY, MULTIPLE PROTEASE FRACTIONS EXHIBITING TRYPTIC AND COLLAGENOLYTIC ACTIVITIES WERE SEPARATED. SOME ENZYMATIC PROPERTIES WERE COMPARED WITH MAJOR PROTEASE FRACTIONS FROM SPINY LOBSTER AND HORSE HAIR CRAB. OPTIMUM PH FOR TRYPTIC ACTIVITY WAS 8.0, WHEREAS THAT FOR COLLAGENOLYTIC ACTIVITY WAS 7.0. ALTHOUGH TRYPTIC ACTIVITY WAS STRONGLY INHIBITED BY DFP, ANTIPAIN, LEUPEPTIN, AND SOYBEAN INHIBITOR, THE INHIBITORY EFFECTS OF THESE AGENTS ON THE COLLAGENOLYTIC ACTIVITY WERE NOT UNIFORM AMONG THE ENZYME PREPARATIONS. THE ENZYME HYDROLYZED -CHAIN OF ACID SOLUBLE COLLAGEN TO FORM -CHAIN WHICH WAS THEN HYDROLYZED TO SMALLER FRAGMENTS. THESE ENZYMES HAVING TRYPTIC ACTIVITY AS WELL AS COLLAGENOLYTIC ACTIVITY MAY CONTRIBUTE TO THE FOOD DIGESTION IN CRUSTACEANS.
