Research Abstract |
We demonstrated that renal growth factor(RGF) was a particular isoform of luteinizing hormone (LH) with microheterogeneity. RGF/IL was purified from porcine pituitary glands, and finally by isoelectric focusing electrophoresis (IEF). IEF separated porcine LH into four fractions based on different pI's. Only IFF-fr.2,pI ranged between 10.28 and 10,18, showed ex vivo renotropic activity. Each IEF fraction showed identical elution profile at reverse-phase HPLC, suggesting that charge heterogeneity among IEF-fractions was due to differences in carbohydrate portion, but not to those in polypeptide chains. Analysis of carbohydrate structure of each subunit, however, failed to show structure specific to IEF-fr.2. Significant in vitro renotropic activity was demonstrated in each IEF-fraction, but their potencies were weaker in fractions with lower pI. These findings duggest that the important role of metabolic clearance rate of each IEF fraction to express in vivo renotropic activity.
