Research Abstract |
1. Germ cell tumors have been reproted to occur in patients with Klinefelter's syndrome. We have made karyotype analysis of the patients with intracranial germc cell tumor to investigate whether the possible genetic factor may be involved in the occurrence of this tumor. Currently, 10 patients (9 males and one female) have been examined. The karyotype was 46XX or 46 XY, and no gross abnormality was observed. We will continue this approach. 2. We have attempted to establish permanent cell lines of intracranial germ cell tumors. A germinoma originated in the suprasellar region in a 17 yearold male has been cultured for 7 months. Number of the cell passage is currently 10. Immunohistochemically, most of the cell is positive for placental alkaline phosphatase and vimentin. Few cells are positive for cytokeratin. Epithelial membrane antigen is negative. This cell is useful to investigate the mechanism of cell differentiation and genetic appraoches for these tumors. 3. Immunihistochemical studies have been performed in the surgical specimen of the intracranial germ cell tumor. No reports have been made by using the antibody against neuron specific enolase (NSE) or Leu 7 in this tumor. All germinomas are diffusely positive for NSE. In embryonal carcinomas, some tumor cells are positive for NSE. Choriocarcinomas are negative for NSE. Leu 7 is positive in some of the large tumor cells in germinomas. Thus, both NSE and Leu 7 are able to be used as tumor markers of the intracranial germ cell tumor.
