Research Abstract

Metabolic changes induced by repetition of epileptic seizure discharges were investigated by comparing the rate of glucose metabolism prior to and after kindling in the same animal through a sequential double-label deoxyglucose (DG) method. The control rate of metabolism prior to kindling was labeled by ^<14>C-DG and the rate in the kindled brain was labeled by ^3H-DG. Each tissue section together with^<14>C and ^3H plastic standards sequentially exposed to an x-ray film and a ^3H sensitive film, respectively. Autoradiographic images were then digitized and exactly overlaid. Based on the difference of sensitivity ratio between the two kinds of films against tracers, optical density equivalent pure ^<14>C and ^3H images were estimated through a digital image processing. Then, metabolic changes were calculated as a (^3H-^<14>C) image, where the distribution of difference values was well fitted by two kinds of normal distributions, ie., one around zero represented a portion which had not changed, and the other around 0.2 represented a portion of increased metabolism after kindling manipulation. Increased metabolism was found in over 60% of the brain investigated, incuding the bilateral dorsal striatum, thalamus (especially VM-VL nuclei), midbrain (red nucleus and substantia nigra), and the dorsal hippocampus.
