Apolipoprotein H (Bata-2-glycoprotein I, abbreviated as apoH) in patients with hyperlipoproteinemia was purified by the step of ultracentrifugation, ion exchange chromatography, heparin Sepharose affinity Chromatography and gel filtration. Polyclonal anti-apoh antibody was prepared by the immunization to rabbits and its gamma-fraction by ammonium persulfate was used for the study. Monoclonal antibody to apoH was also prepared by hybridazation of the spleen cells and mouse myeloma cells (SP-2) after the immunization to mouse (BALB/C). ApoH was able to separate 8 to 10 bands (isoform) after isoelectric focusing. But, after neuraminidase treatment, apoH was converged to three bands. Using SRID (single radial immuno-diffusion) method or latex turbidimetric immunoassay, serum apoH concentration in vaious types of patients were measured. It was clear that serum apoH showed low levels in chronic hepatic hypofunction, high levels during hemodialysis, but normal levels in hyperlipoproteinemia type IIB, IV and Tangier disease. To investigate the releavant of apoH to LDL (low density lipoprotein) incorporated into peripheral cells by LDL-receptor, apoH was afdded in LDL-receptor assay system. ApoH from hyperlipidemic patients inhibited to bind LDL to LDL-receptor.
