Research Abstract

細胞複製にDNAの複製と並んで重要な細胞表層の骨格であるムレイン合成に関与するペニシリン結合蛋白(PBP)と複合体を形成する蛋白を同定し, 抗体を使った次段階に進展した. PBPのプロセス機構と切断部の一次構造が決められ, 高次の構造解析に進んだ. 膜透過に関与するSecY遺伝子とその産物SecYの膜内での配置と膜への組込み機構が明らかとなった. 菌体表面にあるべん毛の遺伝子のオペロン構造を明らかにし, 転写の定量化に成功した. 細菌の接合に関する性フェロモンにより細胞表層に特異的に誘導される蛋白(PD78)の構造解析を行なった. 細胞隔壁形成遺伝子のサプレッサー遺伝子のクローニングが進行し, 細胞膜や隔壁の複製に働く遺伝子発現に理解が深まり次の段階に発展する基礎を完成した.
真核生物の減数分裂前期る誘導されるDNA組換え蛋白の大量精製が進行する一方, invitro遺伝子組換えアッセー系を駆使して分子間と分子内組換え酵素の存在と細胞複製に伴った活性変化を示した. 大腸菌recAに対する抗体と反応するD-loop形成活性をもつ酵素を同定し, これがATP非依存性であることから, 減数分裂細胞にはATP依存型と非依存型の酵素が同時に存在することが示された. 前者は抗recAに反応せず, 後者は抗recAを加えると活性が抑制された. 他方DNA組換え酵素が反応する基質DNAの塩基配列や2次構造と組換え誘導に関する解析をすすめ, 塩基配列だけのような一次構造に対する特異性は存在しないが, 二次構造が重要性をもつことを示す結果を得た. 減数分裂には体細胞周期とは違ったDNA組換え酵素が働くことが多くの物理的・化学的測定で示され, 生殖細胞形成時に特異的な遺伝子発現の切換えが起る機構の解明に前進する基礎ができた. 予定された目的に対する到達度は充分高く, 予期しなかった新発見もあり, 細胞複製と遺伝子発現の相関の解明に確実な進展を示した.
